Recombinant spider silk protein enriched with a fibronectin (RGD) motif for 3D culture of organoids. For correct re-creation of the microenvironment, Biosilk can be biofunctionalised with any full-length human recombinant laminin from BioLamina.
A 3D microfiber network can easily be generated by the gentle introduction of air bubbles into the Biosilk solution. The cell suspension is mixed with Biolaminin and Biosilk. The Biosilk-Biolaminin solution with cells self-assembles into a thin film around each bubble at the air-liquid interface, resembling foam. The bubbles disperse and the foam transforms into a stabilised 3D network with uniformly integrated cells between the microfibers. No cross-linking agent is required.

The mechanical properties will be very much affected by the amount and type of cells included. Biosilk with integrated mesenchymal stem/stromal cells (MSCs) have a Young’s modulus 1.8 ± 0.5 MPa with 25% elongation.
